Cbx8, also known as Pc3, is a member of the Polycomb gene family and is required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. Cbx8 contains a chromodomain and is able to modify chromatin and histones. Function: Component of a Polycomb group (PcG) multiproteinPRC1-like complex, a complex class required to maintain thetranscriptionally repressive state of many genes, including Hoxgenes, throughout development. PcG PRC1 complex acts via chromatinremodeling and modification of histones; it mediatesmonoubiquitination of histone H2A 'Lys-119', rendering chromatinheritably changed in its expressibility. Subunit: Component of a PRC1-like complex. Interacts with RING1RNF2, PCGF1, PCGF2, PCGF3, BMI1, PCGF5 AND PCGF6. Interacts withMLLT3 and histone H3. Interacts with PHC2. Protocol
The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 90% ice-cold methanol for 20 min at-20℃.The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
